Fosta model, Rene Russo avea sa primeasca primul rol dramatic in 1987 in serialul TV Sable, in care a jucat rolul Eden Kendall.
In One Good Cop (1991), joaca rolul unei sotii de politist cu un caracter puternic, a ofiterului Michael Keaton si in 1993 a jucat in Lethal Weapon 3, alaturi de Mel Gibson si Danny Glover.
In In the Line of Fire (1992), Russo a fost din nou partenera unui actor de prima mana, in acest caz agentul secret Clint Eastwood, unul dintre cele mai bune filme.
